以丝素和壳聚糖为原料,采用冷冻干燥法制备丝素/壳聚糖支架,通过预矿化使羟基磷灰石(HA)沉积,形成多孔羟基磷灰石-丝素蛋白/壳聚糖复合支架.对复合支架进行红外吸收光谱(FT-IR)、X射线衍射(XRD)、扫描电子显微镜(SEM)、热重分析(TGA)、力学性能和空隙率表征;采用WST-1方法检测MG-63骨肉瘤细胞在复合支架上的增殖情况,评价其细胞相容性.结果显示,沉积的HA结构属于弱结晶态范畴,主要位于孔壁上,且随着预矿化循环次数的增加,沉积量增多,使预矿化复合支架的弹性模量得到极显著的提高,其孔隙率保持在90%左右的较高水平;WST-1检测表明,含有HA的预矿化复合支架对细胞增殖有明显的促进作用,具有良好的细胞相容性.
