采用Maillard反应制备糖基化蛋白,考察制备方式、共存湿度、缓冲液体系和反应温度、反应时间、pH值等对蛋白接枝度(DG)、褐变化程度(A420)以及改性蛋白乳化稳定性(ES)的影响;利用荧光光谱法考察糖基化蛋白和白蛋白与叶酸作用特征的区别.结果表明,白蛋白与多糖的摩尔比为1:3溶解于pH 8.5Tris-HCl缓冲盐体系,冻干后在RH 79%,55℃条件下、加热反应24h条件下制备得到糖基化蛋白载体.荧光光谱研究结果表明,白蛋白经Maillard反应后,与叶酸的作用增强,作用位点数增加.糖基化蛋白作为疏水性药物栽体,具有提高载药量及靶向的潜力.
